Urochloa brizantha cultivar piatã é uma forrageira tropical amplamente cultivada por seu valor nutritivo e adaptabilidade a solos de baixa fertilidade. Entretanto, o cultivo dessa gramínea apresenta-se ameaçado quando em solos com elevados níveis de sais. A salinidade altera o equilíbrio iônico e o estado redox da planta, provocando alterações que contribuem à perda de produtividade vegetal. Neste sentido, fazem-se necessários métodos para amenizar os efeitos gerados pelo estresse salino, sendo a aplicação de hormônios vegetais uma forma eficiente para tais fins. A proposta do presente estudo é avaliar as respostas de tolerância ao estresse salino em U. brizantha quando tratadas com o ácido indolacético (AIA). Inicialmente foram conduzidos experimentos para selecionar uma concentração adequada do hormônio (50, 200 e 800 µM) via aplicação foliar em plantas submetidas ao estresse salino com NaCl a 100 mM . Foram feitas análises de crescimento pela passagem e medidas de comprimento dos tecidos aéreos e radiculares, determinação dos teores de sódio e potássio por fotômetria de chama, e metodologias espectrofotométricas para análise de conteúdos de clorofilas, carotenoides, prolina e peroxidação lipídica. Os resultados evidenciaram que em menores concentrações de AIA houve uma melhor regulação dos teores de Na + , aumento no conteúdo dos pigmentos fotossintéticos e redução de prolina em plantas estressadas. Sendo assim foi desenvolvido um segundo experimento com a aplicação foliar de 50 µM de AIA quando plantas foram, desta vez, submetidas ao estresse salino com NaCl a 75 mM. Nesse experimento foram avaliados os mesmos parâmetros que no anterior, com a inclusão de análises de conteúdo relativo de água, vazamento de eletrólitos quantificação de peróxido de hidrogênio e atividade das enzimas catalase (CAT), guaiacol peroxidase (POD) e superóxido dismutase (SOD). A atividade enzimática da POD também foi avaliada por zimografia. Os resultados mostraram que o
tratamento com AIA em plantas estressadas não promoveu o crescimento vegetal nem indução de atividade enzimática, entretanto foi constatado a redução do vazamento de eletrólitos, peroxidação lipídica e peróxido de hidrogênio, bem como a diminuição dos teores de sódio e retenção de potássio nas folhas do capim piatã, revelando uma possível participação do AIA na regulação da homeostase iônica. Conclui-se que o AIA induziria a tolerância ao estresse salino em U. brizantha ao agir sobre processos que controlam o transporte de sódio e potássio aos tecidos aéreos.

Urochloa brizantha cultivar piatã is a tropical forage widely cultivated for its nutritive value and adaptability to low fertility soils. However, the growth of this forage grass is threatened when soils contain high levels of salts. Salinity alters the ionic balance and the redox state of the plant, causing alterations that contribute to the loss of plant productivity. In this sense, reliable methods are needed to mitigate the effects generated by saline stress, and the application of plant hormones is an efficient way for such purposes. The aim of this study is to evaluate the salt stress tolerance responses in U. brizantha when treated with indoleacetic acid (IAA). Initially, experiments were carried out to select an adequate concentration of the hormone (50, 200 and 800 µM) via foliar application in plants subjected to saline stress with NaCl at 100 mM. Analysis of growth by weighing and length measurements of aerial and root tissues, determination of sodium and potassium contents by flame photometry, and spectrophotometric methodologies for analysis of chlorophyll, carotenoid, proline and lipid peroxidation contents were carried out. The results showed that at lower concentrations of AIA there was a better regulation of Na+ levels, an increase in the content of photosynthetic pigments and a reduction of proline in stressed plants. Therefore, a second experiment was developed with the foliar application of AIA at 50 µM in plants subjected to saline stress with NaCl at 75 mM. In this experiment, the analyzed parameters in previous experiment were evaluated and it were included the follow analysis: relative water content, electrolyte leakage, content of hydrogen peroxide and enzyme activity of catalase (CAT), guaiacol peroxidase (POD) and superoxide dismutase (SOD). POD enzymatic activity was also evaluated by zymography. The results showed that treatment with AIA in stressed plants did not promote plant growth and stimulate enzyme activity, however it was observed a reduction in electrolyte leakage, lipid peroxidation and hydrogen peroxide, as well as a decrease in sodium levels and potassium retention in piatã grass leaves, revealing a possible participation of AIA in the regulation of ionic homeostasis. It is concluded that AIA would induce salt stress tolerance in U. brizantha by acting on processes that control sodium and potassium transport to the aerial tissues.

Vários fatores podem ocasionar perdas na produção das aliáceas (alho, alho-poró, cebola e cebolinha comum), mas as doenças são muito importantes. Dentre estas destacam-se o tombamento de plântulas, a podridão basal das folhas e nanismo de plantas adultas, causadas por fungos do gênero Rhizoctonia. A identificação desse fungo se baseia, principalmente, na classificação em grupos de anastomose de hifas (AGs). O presente estudo teve como objetivo determinar quais espécies e AGs de Rhizoctonia ocorrem em áreas de cultivo de aliáceas no Brasil usando sequenciamento da região espaçadora transcrita interna (ITS) do DNA ribossômico (rDNA). Plantas de alho, alho-poró, cebola e cebolinha comum com sintomas de tombamento de plântulas, podridão basal das folhas e nanismo de plantas adultas foram coletadas em 38 áreas de produção localizadas em 22 municípios e cinco estados brasileiros, no período de 2018 a 2022. Dos materiais vegetais sintomáticos foram efetuados isolamentos dos fungos associados e obtidas culturas puras de Rhizoctonia. Foram obtidos 49 isolados de Rhizoctonia das áreas de produção de aliáceas. O DNA genômico foi extraído de todos os isolados de Rhizoctonia e efetuada a amplificação por PCR da região ITS-rDNA com os primers ITS1 e ITS4. Os produtos da PCR foram purificados e sequenciados. Sequências nucleotídicas de isolados de referência de Rhizoctonia obtidas no GenBank foram incluídas nas análises e as relações filogenéticas foram analisadas pelo método de Máxima Verossimilhança. A análise filogenética da região ITS permitiu agrupar 44 dos 49 isolados de Rhizoctonia em cinco AGs, sendo três de Rhizoctonia binucleada (AGA-A, AG-F e AG-K) e dois de Rhizoctonia multinucleada, especificamente R. solani (AG-4 HGI e AG-4 HGIII). Cinco isolados foram identificados como Rhizoctonia binucleada mas não se agruparam com nenhum AG descrito. Esse é o primeiro relato, em nível mundial, da associação de AG-F, AG-K e AG- 4 HGI com alho, de AG-A, AG-F e AG-4 HGIII com alho-poró, de AG-F com cebola e de AG-A e AG-F com cebolinha comum. Houve predominância de AG-A (40,8% dos isolados), amplamente distribuído nas áreas brasileiras de produção e associado a todas as espécies de aliáceas aqui estudadas. Isolados representativos dos AGs constatados foram inoculados em cebola e houve diferença significativa nos níveis de agressividade. Esse é o primeiro relato detalhado da composição dos AGs de Rhizoctonia associados com aliáceas no Brasil.

Several factors can cause losses in the production of alliums (garlic, leeks, onions, and chives), but diseases are very important. Among these, the damping-off of seedlings, basal rot of leaves and dwarfing of adult plants, caused by fungi of the genus Rhizoctonia. The identification of this fungus is based mainly on the classification into hyphal anastomosis groups (AGs). The present study was undertaken to determine which Rhizoctonia species and AGs occur in alliums production areas in Brazil using internal transcribed spacer region (ITS) sequencing of ribosomal DNA (rDNA). Garlic, leeks, onions, and chives plants with damping-off of seedlings, basal rot of leaves and dwarfing of adult plants were collected in 38 production areas located in 12 municipalities and five Brazilian states from 2018 to 2022. From symptomatic plant materials were performed isolations of the associated fungi and obtained Rhizoctonia pure cultures. Forty-nine Rhizoctonia isolates were obtained from alliums in Brazil. Genomic DNA was extracted from all Rhizoctonia isolates, and PCR amplifications from the ITS-rDNA region were made with the ITS1 and ITS4 primers. PCR products were purified and sequenced. Nucleotide sequences from Rhizoctonia reference isolates obtained from GenBank were included in the analyses, and phylogenetic relationships were analyzed by Maximum Likelihood method. The phylogenetic analysis of the ITS region allowed grouping 44 of the 49 Rhizoctonia isolates into five AGs, three of binucleate Rhizoctonia (AGA-A, AG- F, and AG-K) and two of multinucleate Rhizoctonia, specifically R. solani (AG- 4 HGI and AG-4 HGIII). Five isolates were identified as binucleate Rhizoctonia but did not cluster with any AG described. This is the first worldwide report of the association of AG-F, AG-K, and AG-4 HGI with garlic, of AG-A, AG-F, and AG-4 HGIII with leek, of AG-F with onion and AG-A and AG-F with chives. There was a predominance of AG-A (40.8% of the isolates), widely distributed in Brazilian production areas and associated with all allium species studied here. Representative isolates of the found AGs were inoculated into onions, and there was a significant difference in aggressiveness levels. This is the first detailed report on the composition of Rhizoctonia AGs associated with alliums in Brazil.

Lectinas são proteínas bioativas naturais encontradas em bactérias, fungos, animais e plantas, capazes de interagir com oligossacarídeos, monossacarídeos e glicoconjugados por meio de um domínio de reconhecimento a carboidratos. Lectinas de plantas, especialmente as das leguminosas, têm sido extensivamente estudadas por apresentarem variados efeitos biológicos, incluindo a capacidade de inibir o crescimento de fungos. Em virtude de sua atividade fungicida e da biodegradabilidade, lectinas vegetais podem ser empregadas no controle das doenças causadas por fungos fitopatogênicos. Diante disso, este trabalho buscou avaliar o efeito inibitório das lectinas de Vatairea macrocarpa (VML) e de Parkia platycephala (PPL) sobre o crescimento dos fungos fitopatogênicos Colletotrichum okinawense, C. siamense, Lasiodiplodia theobromae e Rhizoctonia solani AG-4 HGI. Para tanto, as lectinas foram purificadas por cromatografia de afinidade, liofilizadas e testadas quanto à pureza por SDS-PAGE e à atividade hemaglutinante. Para testar o efeito inibitório das lectinas, os isolados fúngicos foram cultivados em meio BDA contendo VML (1000 μg/mL) e PPL (100 e 200 μg/mL), separadamente. Como controles positivos, foram usados os fungicidas pencicurom e tiofanato metílico. Placas contendo somente BDA foram utilizadas como testemunhas. O crescimento de C. siamense (isolado CMM 4081) reduziu com a utilização de tiofanato metílico e da lectina PPL a 200 μg/mL, sem diferirem entre si, mas diferindo dos demais tratamentos. Na presença da lectina PPL a 200 μg/mL, o crescimento do isolado CMM 2295 de L. theobromae foi inferior ao observado na testemunha. A lectina VML foi efetiva na redução do crescimento micelial do isolado CMM 4039 de L. theobromae, assemelhando-se ao observado com a utilização de tiofanato metílico. As lectinas VML e PPL não apresentaram atividade inibitória sobre o crescimento de C. okinawense e R. solani. A atividade inibitória dessas lectinas pode ser espécie-dependente em relação a Colletotrichum e isolado-dependente em relação a L. theobromae. Os resultados estimulam futuras pesquisas com lectinas visando a obtenção de formulações para o controle das doenças causadas por fungos em plantas na pós-colheita.

Lectins are natural bioactive proteins found in bacteria, fungi, animals, and plants, capable of interacting with oligosaccharides, monosaccharides, and glycoconjugates through a carbohydrate recognition domain. Plant lectins, especially those from legumes, have been extensively studied because they have various biological effects, including the ability to inhibit fungal growth. Due to their fungicidal activity and biodegradability, plant lectins can be used to control diseases caused by phytopathogenic fungi. Therefore, this work sought to evaluate the inhibitory effect of lectins from Vatairea macrocarpa (VML) and Parkia platycephala (PPL) on the growth of phytopathogenic fungi Colletotrichum okinawense, C. siamense, Lasiodiplodia theobromae and Rhizoctonia solani AG-4 HGI. Therefore, the lectins were purified by affinity chromatography, lyophilized, and tested for purity by SDS-PAGE and hemagglutinating activity. To test the inhibitory effect of lectins, fungal isolates were cultured in PDA medium containing VML (1000 μg/mL) and PPL (100 and 200 μg/mL), separately. As positive controls, pencycuron and methyl thiophanate fungicides were used. Plates containing only BDA were used as controls. The growth of C. siamense (isolate CMM 4081) reduced with the use of methyl thiophanate and PPL lectin at 200 μg/mL, without differing from each other, but differing from the other treatments. In the presence of the PPL lectin at 200 μg/mL, the growth of the L. theobromae isolate CMM 2295 was lower than that observed in the control. The VML lectin effectively reduced the mycelial growth of the L. theobromae isolate CMM 4039, similar to that observed with methyl thiophanate. The VML and PPL lectins did not show inhibitory activity on the growth of C. okinawense and R. solani. The inhibitory activity of these lectins can be species-dependent regarding Colletotrichum and isolated-dependent in relation to L. theobromae. The results encourage future research with lectins aimed at obtaining formulations to control diseases caused by fungi in post-harvest plants.

As leishmanioses são doenças vetoriais de grande impacto no cenário da saúde pública, estando presente em mais de 98 países, sendo responsáveis pela exposição de aproximadamente 350 milhões de pessoas. Os vetores exercem importante papel no ciclo biológico da doença. Dada a importância dessa endemia, em particular da forma cutânea, os aspectos entomológicos dos flebotomíneos transmissores da Leishmaniose Tegumentar Americana (LTA) e o agente etiológico foram estudados no município de Barbalha – CE. Durante o período de janeiro e dezembro de 2022 foram realizadas coletas de amostras das lesões de pacientes atendidos no Ambulatório de Especialidades Médicas da Faculdade de Medicina da Universidade Federal do Cariri e coletas sistematizadas dos flebotomíneos no domicílio e peridomicílio desses pacientes, sendo avaliadas e classificadas as espécies dos insetos vetores, complementado por análises moleculares direcionadas ao gênero Leishmania spp.. A captura dos flebotomíneos foi realizada com armadilhas luminosas CDC (Centers for Disease Control), instaladas no intradomicílio e peridomicílio dos pacientes diagnosticados com LTA. Os vetores coletados foram triados e posteriormente classificados de acordo com o sexo e a espécie. Foram coletados 1091 flebotomíneos, sendo 510 (46,7%) machos e 581 (53,3%) fêmeas, com 11 espécies classificadas do gênero Lutzomyia, assim discriminadas: L. longipalpis (15%), L. sallesi (0,7%), L. lenti (6,8 %), L. sordellii (0,3 %), L. intermedia (39,1 %), L. quinquefer (1,4%), L. goiana (1,9%), L. whitmani (30,5%), L. evandroi (0,3%), L. migonei (1,0%) e L. walqueri (3%). Os dados obtidos permitem concluir que a fauna flebotômica em Barbalha é diversa e difere em alguns padrões dos dados das demais região do estado do Ceará e que estudo complementar para caracterizar a(s) espécie(s) envolvidas na transmissão complementará o presente ensaio.

Leishmaniasis are vector diseases that have a major impact on the public health scenario, being present in more than 98 countries and responsible for the exposure of approximately 350 million people. Vectors play an important role in the biological cycle of the disease. Given the importance of this endemic, particularly the cutaneous form, the entomological aspects of the sandflies that transmit American Cutaneous Leishmaniasis (ACL) and the etiological agent were studied in the municipality of Barbalha – CE. During the period of January and December 2022, samples were collected from lesions of patients treated at the Medical Specialties Outpatient Clinic of the Faculty of Medicine of the Federal University of Cariri and systematic collections of sandflies were carried out in the homes and surroundings of these patients, evaluating and classifying the species of vector insects, complemented by molecular analyzes directed to the genus Leishmania spp.. The capture of sandflies was carried out using CDC (Centers for Disease Control) light traps, installed in the intra-domestic and non-domestic homes of patients diagnosed with ACL. The collected vectors were screened and subsequently classified according to sex and species. A total of 1091 sandflies were collected, 510 (46.7%) males and 581 (53.3%) females, with 11 species classified as the genus Lutzomyia spp., broken down as follows: L. longipalpis (15%), L. sallesi (0.7 %), L. lenti (6.8 %), L. sordellii (0.3 %), L. intermedia (39.1 %), L. quinquefer (1.4%), L. goiana (1.9 %), L. whitmani (30.5%), L. evandroi (0.3%), L. migonei (1.0%) and L. walqueri (3%). The data obtained allow us to conclude that the phlebotomy fauna in Barbalha is diverse and differs in some patterns from data from other regions of the Ceará state and that a complementary study to characterize the species(s) involved in transmission of ACL will complement the present essay.

A azitromicina (AZT) está entre os antibióticos mais bem-sucedidos e altamente prescritos no mundo, sendo utilizada no tratamento de primeira linha para infecções do trato respiratório, doenças sexualmente transmissíveis, infecções dérmicas e outras manifestações bacterianas. Entretanto, para os casos em que se faz necessário medir os níveis do princípio ativo em matrizes complexas, ou ainda, quando se quer estudar a reatividade e estabilidade da molécula, o número de possibilidades analíticas com os requisitos necessários para estas finalidades ainda é limitado. Neste trabalho, uma proposta inovadora foi desenvolvido com um sensor eletroquímico à base de molibdato de cério (Ce2(MoO4)3) e nanotubos de carbono de parede múltiplas (MWCNT) integrados em uma pasta condutora de carbono. Apesar da elevada reatividade dos materiais presentes no dispositivo resultante (Ce2(MoO4)3-MWCNT/EPC) para analisar a oxidação do antibiótico, dados experimentais e simulações computacionais revelaram que a solubilidade e eletroatividade do antibiótico melhoram em eletrólito alcalino (pH = 8) preparado em mistura binária de CH3OH/H2O (10:90%, v/v). A resistência de transferência de carga e, consequentemente, a sensibilidade do dispositivo também melhorou após o tratamento térmico de Ce2(MoO4)3-MWCNT/EPC, como forma de eliminar a água reticular contida nos microcristais de molibdato. Há evidências de que a oxidação do antibiótico sobre o sensor eletroquímico ocorre de forma irreversível, com indícios de liberação de 2e‒ da desosamina. Após a otimização dos parâmetros eletroanalíticos, foi possível detectar AZT com limite de detecção de 230 nm, além de alta precisão dos resultados, demonstrada por desvios de resultados abaixo de 4,5%. O dispositivo mostrou aplicabilidade para avaliar a estabilidade termo-oxidativa da AZT sob condições variadas de temperatura, radiação ultravioleta e saturação ao ar atmosférico. Os resultados fortalecem a ideia de que o controle de qualidade dos medicamentos à base de AZT deve ser feito de forma contínua, em padrões laboratoriais não-ideais e mais próximos daqueles observados durante o transporte, armazenamento e administração dos produtos, como forma de fazer uma avaliação mais rigoroso da validade e viabilidade do ingrediente ativo nas formulações, assim como zelar pela saúde dos consumidores.

Azithromycin (AZT) is among the most successful and highly prescribed antibiotics in the world, being used in first-line treatment for respiratory tract infections, sexually transmitted diseases, dermal infections, and other bacterial manifestations. However, when it is necessary to measure the levels of this active ingredient in complex matrices, or even when you want to study the reactivity and stability of the molecule, the number of analytical possibilities with the necessary requirements for these purposes is still limited. In this work, an innovative proposal was developed with an electrochemical sensor based on cerium molybdate (Ce2(MoO4)3) and multi-walled carbon nanotubes (MWCNT) integrated into a conductive carbon paste. Despite the high reactivity of the materials present in the resulting device (Ce2(MoO4)3-MWCNT/EPC) to analyze the oxidation of the antibiotic, experimental data and computational simulations revealed that the solubility and electroactivity of the antibiotic improve in alkaline electrolyte (pH = 8) prepared in a binary mixture of CH3OH/H2O (10:90%, v/v). The charge transfer resistance and, consequently, the sensitivity of the device also improved after the heat treatment of Ce2(MoO4)3-MWCNT/EPC, as a way to eliminate the reticular water contained in the molybdate microcrystals. There is evidence that the oxidation of the antibiotic on the electrochemical sensor occurs irreversibly, losing 2e‒ of desosamine. After optimizing the electroanalytical parameters, it was possible to detect AZT with a detection limit of 230 nm, in addition to high precision of the results, demonstrated by results deviations below 4.5%. The device showed applicability to evaluate the thermo-oxidative stability of AZT under varying conditions of temperature, ultraviolet radiation and atmospheric air saturation. The results strengthened the idea that quality control of AZT-based medicines must be carried out continuously, using non-ideal laboratory standards and closer to those observed during transport, storage and administration of the products, as a way of making a more rigorous assessment of the validity and viability of the active ingredient in formulations, as well as ensuring the health of consumers.

A hipertrofia cardíaca é caracterizada pelo aumento do tamanho do coração decorrente de estímulos intracelulares adquiridos de uma sobrecarga de pressão ou volume. Embora seja uma resposta adaptativa e transitória, frequentemente evolui para um quadro de insuficiência cardíaca congestiva, ocasionando falha cardíaca. Disfunções mitocondriais estão envolvidas no desenvolvimento de patologias cardíacas e que geralmente são associadas com defeitos na cadeia de transporte de elétrons, sobrecarga de Ca2+, reativação de genes cardíacos e principalmente, pelo aumento de estresse oxidativo. Por outro lado, a restrição calórica (RC) é uma intervenção nutricional que protege contra a hipertrofia cardíaca por diminuição do estresse oxidativo e melhora da função mitocondrial. Neste estudo, será mostrado que esta intervenção dietética previne a elevação das proteínas cardíacas, evita a reprogramação do fator natriurético atrial e bloqueia o aumento do índice de peso do coração por comprimento da tíbia (mg/cm), parâmetros observados em corações hipertróficos. Esses resultados sugerem que a RC inibe o crescimento patológico cardíaco, ao mesmo tempo em que reduz a formação de peróxido de hidrogênio induzida pelo transporte reverso de elétrons mitocondrial e melhora o conteúdo mitocondrial. Adicionalmente, a RC atenuou a abertura do mPTP induzida por Ca2+ em mitocôndrias isoladas de camundongos tratados com isoproterenol. Além disso, a RC bloqueou a correlação negativa de enzimas antioxidantes (atividade de superóxido dismutase e glutationa peroxidase), levando à manutenção dos níveis de proteína sulfidrilas e glutationa. Dada a natureza da hipertrofia cardíaca induzida por isoproterenol, foi investigado se a RC pode alterar a sensibilidade β-adrenérgica cardíaca. Assim, usando corações de ratos isolados em um sistema de Langendorff, foi visto que ratos com RC (semelhante aos controles) preservaram a sinalização β-adrenérgica. Em contrapartida, corações de ratos hipertróficos (tratados por sete dias com isoproterenol) foram insensíveis à ativação β-adrenérgica com isoproterenol (50 nM). Esses resultados indicam que a RC possui efeitos benéficos contra a cardiomiopatia hipertrófica e podem contribuir para prevenção e/ou tratamento de complicações relacionadas a hipertrofia cardíaca e outras doenças cardiovasculares.

Cardiac hypertrophy is characterized by an increase in heart size due to intracellular stimuli acquired from pressure or volume overload. Although it is an adaptive and transient response, it often evolves into congestive heart failure, causing heart failure. Mitochondrial dysfunctions are involved in the development of cardiac pathologies and are generally associated with defects in the electron transport chain, Ca2+ overload, reactivation of cardiac genes and, mainly, increased oxidative stress. On the other hand, calorie restriction (CR) is a nutritional intervention that protects against cardiac hypertrophy by decreasing oxidative stress and improving mitochondrial function. In this study, it will be shown that this dietary intervention prevents the elevation of cardiac proteins, avoids the reprogramming of the atrial natriuretic factor and blocks the increase in the index of heart weight per length of the tibia (mg/cm), parameters observed in hypertrophic hearts. These results suggest that CR inhibits pathological cardiac growth while reducing mitochondrial reverse electron transport-induced hydrogen peroxide formation and improving mitochondrial content. Additionally, RC attenuated Ca2+-induced mPTP opening in mitochondria isolated from mice treated
with isoproterenol. In addition, CR blocked the negative correlation of antioxidant enzymes (superoxide dismutase and glutathione peroxidase activity), leading to the maintenance of sulfhydryl and glutathione protein levels. Given the nature of isoproterenol-induced cardiac hypertrophy, we investigated whether CR could alter cardiac β-adrenergic sensitivity. Thus, using isolated rat hearts in a Langendorff system, it was seen that rats with CR (similar to controls) preserved β-adrenergic signaling. In contrast, hypertrophic rat hearts (treated for seven days with isoproterenol) were insensitive to β-adrenergic activation with isoproterenol (50 nM). These results indicate that CR has beneficial effects against hypertrophic cardiomyopathy and may contribute to the prevention and/or treatment of complications related to cardiac hypertrophy and other cardiovascular diseases.

Os carboidratos são as moléculas orgânicas mais abundantes do planeta, sendo importantes em mecanismos de reconhecimento e interação intercelular e para a saúde. Diferentes formas de controle dos níveis de glicose tem sido alvo de muitas pesquisas que visam facilitar e/ou aprimorar a detecção desse monossacarídeo em fluidos biológicos, alimentos e formulações comerciais. Devido à capacidade de ligação reversível e específica a carboidratos, as lectinas são proteínas que vêm ganhando espaço como moléculas de reconhecimento no desenvolvimento de biossensores eletroquímicos para glicose. As lectinas de leguminosa são amplamente estudadas e caracterizadas, sendo a lectina isolada de Canavalia ensiformes (ConA) a mais conhecida desse grupo. Pela diversidade de propriedades de acordo com a fonte de obtenção, as lectinas de fungos também mostram potencial biotecnológico. A espécie Ganoderma applanatum, por exemplo, apresenta uma lectina (GAL) com afinidade por glicose e que apresenta resistência moderada a variações de pH (4 - 8) e temperatura (20 – 60 ºC), embora com atividade hemaglutinante melhor conservada em condições fisiológicas. Neste trabalho foram desenvolvidos dois sistemas de biorreconhecimento para glicose com a ConA e a GAL, utilizando eletrodo de carbono vítreo (GCE) modificado com o hexacianometalato Azul da Prússia (PB) que, além de ser um material semicondutor, possui regiões de densidade eletrônica heterogêneas e adequadas para a adsorção das proteínas, através de ligações intermoleculares. Os resultados obtidos por diferentes técnicas comprovaram que o problema de instabilidade do PB em pH fisiológico pode ser solucionado por tratamento térmico a 100 ºC por 1h, levando ao suporte eletródico de ta-PB/GCE. O uso de inteligência artificial associado a parâmetros eletroquímicos possibilitou estudos estruturais e a obtenção de dados sobre as condições biocompatíveis e eletrostaticamente favoráveis para imobilização da GAL em ta-PB/GCE, que também foram úteis nos estudos com a ConA. O biossensor GAL/ta-PB/GCE apresentou ótimo desempenho eletroquímico para a quantificação de glicose em condições ideais, alcançando um limite de detecção de 10,2 pM e sensibilidade de 0,012 µA µM ‒1 cm ‒2 . A performance obtida com ConA/ta-PB/GCE foi satisfatória, alcançando um limite de detecção de 0,96 µM e sensibilidade de 3,94 × 10 3 µA µM ‒1 cm ‒2 . A seletividade dos ensaios foi influenciada por outros carboidratos, incluindo frutose, maltose e sacarose, provavelmente pela regiosseletividade moderadamente baixa alcançada com sítios de interação superficiais dessas proteínas, mas sem comprometer a viabilidade dos dispositivos para a eletroanálise de glicose em amostras complexas. Ambos os biossensores foram testados para a quantificação de glicose em formulações farmacêuticas, alcançando valores de recuperação entre 98,0% e 100,5%. O biossensor ConA/ta-PB/GCE foi aplicado com êxito para a análise de açúcares totais em amostra de refrigerante. Cada dispositivo apresentou vantagens particulares e desafios que ainda demandam mais estudos, porém o conjunto de figuras de mérito obtido em cada sistema comprovou que as plataformas desenvolvidas são verdadeiramente eficientes para a análise de glicose em diferentes amostras, estimulando mais pesquisas com o desenvolvimento de biossensores eletroquímicos à base de lectinas para carboidratos de relevância biológica.

Carbohydrates are the most abundant organic molecules on the planet, being important in mechanisms of intercellular recognition and interaction and for health. Different ways of controlling glucose levels have been the subject of much research aimed at facilitating and/or improving the detection of this monosaccharide in biological fluids, foods and commercial formulations. Due to their ability to reversibly and specifically bind carbohydrates, lectins are proteins that have been gaining ground as a recognition star in the development of electrochemical glucose biosensors. Legume lectins are widely studied and characterized, with the basic lectin from Canavalia ensiformes (ConA) being the best-known of this group. Due to the diversity of properties according to the source of production, fungal lectins also show biotechnological potential. The species Ganoderma applanatum, for example, has a lectin (GAL) with an affinity for glucose and which is moderately resistant to variations in pH (4 - 8) and temperature (20 – 60 ºC), although with a hemagglutinating activity that is better preserved under physiological conditions. In this work, two biorecognition systems for glucose were developed with ConA and GAL, using a glassy carbon electrode (GCE) modified with hexacyanometalate Prussian Blue (PB) which, in addition to being a semiconductor material, has heterogeneous electron density regions and suitable for the adsorption of proteins, through intermolecular bonds. The results obtained by different techniques proved that the problem of PB instability at physiological pH can be solved by thermal treatment at 100 ºC for 1h, leading to the electrode support of ta-PB/GCE. The use of artificial intelligence associated with electrochemical parameters allowed structural studies and the collection of data on biocompatible and electrostatically favorable conditions for immobilization of GAL in ta-PB/GCE, which were also useful in studies with ConA. The GAL/ta-PB/GCE biosensor showed excellent electrochemical performance for glucose quantification under ideal conditions, reaching a detection limit of 10.2 pM and sensitivity of 0.012 µA µM ‒1 cm ‒2 . The performance obtained with ConA/ta-PB/GCE was satisfactory, reaching a detection limit of 0.96 µM and sensitivity of 3.94×10 3 µA µM ‒1 cm ‒2 . The selectivity of the assays was influenced by other carbohydrates, including fructose, maltose and sucrose, probably due to the moderately low regioselectivity achieved with surface interaction sites of these proteins, but without compromising the viability of the devices for the electroanalysis of glucose in complex samples. Both biosensors were tested for glucose quantification in pharmaceutical formulations, achieving recovery values between 98.0% and
100.5%. The ConA/ta-PB/GCE biosensor was successfully applied for the analysis of total sugars in a soft drink sample. Each device presented particular advantages and challenges that still require further studies, but the set of figures of merit obtained in each system proved that the platforms developed are truly efficient for the analysis of glucose in different samples, stimulating further research with the development of electrochemical biosensors based on lectins for carbohydrates of biological relevance.

A hipertrofia cardíaca é uma resposta compensatória do coração frente a um aumento de volume ou pressão arterial. Embora seja inicialmente uma resposta benéfica, frequentemente, leva à cardiomiopatia hipertrófica e à insuficiência cardíaca. Estudos apontam que a hipertrofia cardíaca induz um shift metabólico levando a um consumo preferencial de glicose em relação aos ácidos graxos. A restrição calórica, um procedimento dietético que promove benefícios à saúde e prolonga a expectativa de vida em muitos organismos, produz efeitos que envolvem melhora no metabolismo energético e estresse oxidativo, no qual as mitocôndrias desempenham um papel central. Neste trabalho, levantou-se a hipótese de que parte dos efeitos benéficos da restrição calórica se dá por alterações no conteúdo lipídico, manutenção das funções mitocondriais e redox, e que somados, estes efeitos induziriam à prevenção da hipertrofia cardíaca. A fim de investigá-los, um protocolo de indução da restrição calórica em camundongos foi realizado com base no consumo médio da dieta padrão de camundongos alimentados ad libitum durante 2 semanas. Em seguida, os animais foram divididos, aleatoriamente, em dois grupos: com ingestão ad libitum (AL), e alimentado com 60% do consumo médio de calorias (RC) durante 4 semanas adicionais. A hipertrofia cardíaca foi induzida por isoproterenol (ISO) (30 mg/ kg / dia) durante 8 dias. Camundongos tratados com ISO tiveram elevada relação peso do coração/ comprimento da tíbia e aumento da área transversal dos cardiomiócitos. Esses marcadores hipertróficos foram significantemente reduzidos em camundongos RC tratados com ISO. Mitocôndrias isoladas de corações hipertróficos produziram níveis significativamente mais altos de H2O2 (usando succinato na presença de rotenona). Este efeito foi bloqueado pela RC. Concomitantemente a hipertrofia cardíaca inibiu a atividade das enzimas, MnSOD, GPx, e GST, reduziu as taxas de controle respiratório mitocondrial e diminuiu a atividade ATPase . Esses efeitos também foram evitados pela RC. Para obter informações sobre como a RC pode interferir nas alterações metabólicas induzidas pela hipertrofia, o lipidoma cardíaco também foi estudado. A RC protegeu contra mudanças em vários triglicerídeos (TAGs) ligados a ácidos graxos insaturado, além de proteger contra o acúmulo de TAGs contendo ácidos graxos saturados observados em amostras hipertróficas. A hipertrofia cardíaca induziu aumento de ceramidas, fosfoetanolaminas e acilcarnitinas (12:0, 14:0, 16:0 e 18:0), e promoveu dano mitocondrial observado pelo inchamento secundário à absorção de Cálcio. Estas alterações foram evitadas pela restrição calórica. Juntos, estes dados demonstram que a hipertrofia promove o estresse oxidativo, distúrbios mitocondriais, e altera o lipidoma cardíaco. Essas alterações são evitadas (pelo menos parcialmente) pela intervenção de restrição calórica in vivo. Essas descobertas podem ter implicações promissoras no desenvolvimento de novas estratégias de tratamento para proteger o coração contra a hipertrofia e suas complicações. No entanto, mais pesquisas são necessárias para aprofundar nosso entendimento sobre os mecanismos exatos pelos quais a restrição calórica exerce seus efeitos benéficos no coração.

Cardiac hypertrophy is a compensatory response of the heart to an increase in volume or blood pressure. Although initially a beneficial response, it often leads to hypertrophic cardiomyopathy and heart failure. Studies indicate that cardiac hypertrophy induces a metabolic shift leading to a preferential consumption of glucose in relation to fatty acids. Caloric restriction, a dietary procedure that promotes health benefits and prolongs life expectancy in many organisms, produces effects involving improvement in energy metabolism and oxidative stress, in which mitochondria play a central role. In this work, the hypothesis was raised that part of the beneficial effects of caloric restriction is due to changes in lipid content, maintenance of mitochondrial and redox functions, and that together, these effects would induce the prevention of cardiac hypertrophy. To investigate these effects, a protocol for the induction of caloric restriction in mice was performed based on the average consumption of the standard diet of mice fed ad libitum for 2 weeks. The animals were then randomly divided into two groups: ad libitum intake (AL) and fed 60% of the mean calorie intake (CR) for an additional 4 weeks. Cardiac hypertrophy was induced by isoproterenol (ISO) (30 mg/kg/day) for 8 days. Mice treated with ISO had an increased heart weight/tibial length ratio and increased cardiomyocyte cross-sectional area. These hypertrophic markers were significantly reduced in ISO-treated CR mice. Mitochondria isolated from hypertrophic hearts produced significantly higher levels of H2O2 (when succinate was used without the presence of rotenone). This effect was blocked by CR. Concomitantly, cardiac hypertrophy inhibited the activity of enzymes, MnSOD, GPx, and GST, reduced rates of mitochondrial respiratory control and decreased the ATPase activity. These effects were also avoided by CR. To obtain information on how CR can interfere with hypertrophy-induced metabolic changes, cardiac lipidome was also studied. CR protected against changes in several triglycerides containing saturated fatty acids observed in hypertrophic samples. Cardiac hypertrophy induced an increase in ceramides, phosphoethanolamines and acylcarnitines (12:0, 14:0, 16:0 and 18:0), and promoted mitochondrial damage observed by swelling secondary to calcium absorption. These changes were prevented by caloric restriction. Together, these data demonstrate that hypertrophy promotes oxidative stress, mitochondrial disorders, and alters cardiac lipidome. These changes are prevented (at least partially) by in vivo caloric restriction intervention. These findings could have promising implications for the development of new treatment strategies to protect the heart against hypertrophy and its complications. However, more research is needed to deepen our understanding of the exact mechanisms by which calorie restriction exerts its beneficial effects on the heart.
